The Medicinal Chemistry Shared Resource - High Throughput Screening Core (MCSR-HTSC) specializes in biochemical and cell-based in vitro assays using high throughput and automation-friendly microplate formats. We place a strong focus on the physiological relevance of plate-based assays and employ a range of models that include 2D and 3D mono-culture and co-cultures, including the use of both spheroids and organoids. These models can be used for fixed (e.g., cell painting) and live cell studies, leveraging techniques that include high-content confocal imaging and high throughput flow cytometry on 384-well plates. Additionally, we are investigating human iPSC-derived models in cytotoxicity and functional assays. We have successfully established a comprehensive early drug discovery workflow encompassing hit generation, characterization, and mechanistic exploration underpinned by integrated data, image analysis, and management systems. We will showcase several screening initiatives demonstrating this core facility's expertise, which serves as a pivotal resource at Ohio State University for initiating both hypothesis-driven and unbiased, systematic, scalable early drug discovery endeavors.
